Activation of Crude a N D Homogeneous Juvenile Hormone Esterases by Organic Solvents
نویسنده
چکیده
-A variety of water-miscible organic solvents caused profound activation of the homogeneous juvenile hormone esterase from Manduca sexta. Much lower activation was observed with other species. Methods for the handling of solutions of juvenile hormone are presented. The implications of solvent activation of juvenile hormone esterase activity are discussed, especially as they relate to analysis of the enzyme or titre determinations of juvenile hormone.

Increasing Performance and Thermostability of D-Phenylglycine Aminotransferase in Miscible Organic Solvents
Background: D-Phenylglycine aminotransferase (D-PhgAT) is highly beneficial in pharmaceutical biotechnology. Like many other enzymes, D-PhgAT suffers from low stability under harsh processing conditions, poor solubility of substrate, products and occasional microbial contamination. Incorporation of miscible organic solvents into the enzyme’s reaction is considered as a solution...
متن کاملUse of Cross-Linked Enzyme Aggregates (CLEAs) for performing biotransformations
The methodology for the preparation of cross-linked enzyme aggregates (CLEAs) and their application as highly effective, recyclable biocatalysts in a wide variety of transformations is reviewed. The method constitutes a combination of enzyme purification and immobilization into a single operation and allows for the use of relatively crude enzyme preparations. The resulting CLEAs have excellent ...
متن کاملJuvenile hormone (JH) esterase: why are you so JH specific?
Juvenile hormone esterases (JHEs) from six insects belonging to three orders (Lepidoptera, Coleoptera, and Diptera) were compared in terms of their deduced amino acid sequence and biochemical properties. The four lepidopteran JHEs showed from 52% to 59% identity to each other and about 30% identity to the coleopteran and dipteran JHEs. The JHE of Manduca sexta was remarkably resistant to the ad...
متن کاملIn vitro expression and biochemical characterization of juvenile hormone esterase from Manduca sexta.
Juvenile hormone esterase (JHE) is a selective enzyme that hydrolyzes the methyl ester of juvenile hormone. This enzyme plays an important role in the regulation of metamorphosis in caterpillars, and is implicated in additional roles in development and reproduction in this and other orders of insect. The full length coding region of the JHE cDNA from Manduca sexta was subcloned into the baculov...
متن کاملEsterase Active in Polar Organic Solvents from the Yeast Pseudozyma sp. NII 08165
Esterases/lipases active in water miscible solvents are highly desired in biocatalysis where substrate solubility is limited and also when the solvent is desired as an acyl acceptor in transesterification reactions, as with the case of biodiesel production. We have isolated an esterase from the glycolipid producing yeast-Pseudozyma sp. NII 08165 which in its crude form was alkali active, thermo...
